In vitro symbiotic seed germination in Vanda wightii, an endemic orchid species of Western Ghats, India supported by Ceratobasidiaceae isolates

Symbiotic seed germination for conservation and cultivation of orchids holds colossal merit asmycorrhizal fungus in its system improves their growth adaptability. activity ishighly specific some species, but other cases the same from one species is effective aseries related species. The present work describes inter-specific three fungal isolatesfrom seedling root Vanda thwaitesii to support V.wightii, a closely India. Among isolates, two designated as Wyd2 andIdk were identified Ceratobasidium sp through sequencing ITS1 ITS4 regions. Oneisolate Wyd1 did not clad with any described genera, remained an out-groupunder family Ceratobasidiaceae. All isolates possessed binucleate hyphae producingellipsoidal, oval or barrel shaped monilioid cells supported 80–95% germination,transforming 70–85% them into protocorms 30 days duration. seedlingdevelopment starting promeristem formation, first leaf development second leafinitiation occurred 95% time interval 60 compared 90–120 daysthrough asymbiotic method. evaluated are provedeffective V. wightii symbiotic thus useful mycorrhiza assistedconservation.
